Event-specific qualitative polymerase chain reaction analysis for two T-DNA copies in genetically modified orange Petunia
In 2017, a variety of orange-colored petunias in the market turned out to be genetically modified (GM) without official authorization for commercialization. sequence analysis suggested these plants declared most probably derived from the plant transformation experiments done in the 1980s. For a deeper understanding of how GM petunias entry classical breeding programs around the world, and whether they came from a single source or not, we do molecular genetic characterization of T-DNA integration sites in a variety of GM petunia cultivars and breeding lines.
By way of running the genome, we isolated a T-DNA sequence is different, which is located at the junction between the T-DNA (s) and petunia DNA. Based on the results we conclude that there are at least two copies of the T-DNA of different lengths. This is supported by Southern blot analysis. For T-DNA1, 3′-junction sequence is isolated, while the 5′-junction is still unclear.
Conversely, for T-DNA2, the sequence 5′-junction isolated, while the sequence is isolated from the 3′-region consists only of the T-DNA, but does not include the intersection of the T-DNA into the DNA petunias. We developed a PCR primer for a specific event and screened a set of three orange petunia cultivars GM and GM 126 offspring from a commercial breeding program. We show that the second copy of the T-DNA that is present in all tested samples of petunias our GM, which supports the assumption of a single transgenic origin of petunias GM declared. Most likely, two T-DNA integrated into the genome petunia close.

Primer pairs targeting conserved regions PPV6 capsid gene was designed. sensitivity analysis revealed lower limit of detection of real-time PCR based on SYBR Green to 47.8 copies / uL, indicating it is 1000 times higher than that found in conventional PCR probe.

Positive detection rate of 180 clinical samples with SYBR Green-based real-time PCR and conventional PCR was 12.22% (22/180) and 4.44% (8/180), respectively. Our method is sensitive, specific, and reproducible. The use of SYBR Green-based real-time PCR detection may be suitable for clinical and epidemiological investigation of PPV6.
